Chemists have now unravelled this decade-old mystery, in the process revealing that the dark blue pigments at the centre of the mystery are similar to&nbsp;<a href=\"https:\/\/www.chemistryworld.com\/3009550.article\">indigo<\/a>, the dye used to produce blue jeans.<\/p>\n\n\n\n<p>Magic mushrooms or\u00a0<em>Psilocybe<\/em>\u00a0are fungi producing the psychotropic compounds\u00a0<a href=\"https:\/\/trufflemushroomcenter.com\/\">psilocybin<\/a>\u00a0and psilocin. They are one of several species that instantly develop a blue coloration when they are cut or bruised. In\u00a0<a href=\"https:\/\/en.wikipedia.org\/wiki\/Gyroporus_cyanescens\"><em>Boletales<\/em><\/a>\u00a0mushrooms, oxidised gyrocyanin or pulvinic acid are the source of the blue colour. But that isn\u2019t the case in\u00a0<em>Psilocybe\u00a0<\/em>mushrooms.<\/p>\n\n\n\n<p>Previous research had established that the blue colour was caused by oxidised psilocybin, but the nature of the pigment and the biochemical pathway producing it had remained elusive.<\/p>\n\n\n\n<figure class=\"wp-block-image\"><a href=\"https:\/\/d2cbg94ubxgsnp.cloudfront.net\/Pictures\/\/web\/w\/v\/r\/lenzetalscheme1_974318.svgz\"><img decoding=\"async\" src=\"https:\/\/d2cbg94ubxgsnp.cloudfront.net\/Pictures\/\/web\/w\/v\/r\/lenzetalscheme1_974318.svgz\" alt=\"An image showing the chemical structures of psilocybin and psilocin\"\/><\/a><\/figure>\n\n\n\n<p>Magic mushrooms contain a number of psychotropic compounds including psilocybin and psilocin.<\/p>\n\n\n\n<p><a href=\"https:\/\/www.leibniz-hki.de\/en\/institut-staff-details.html?member=73\">Dirk Hoffmeister<\/a>&nbsp;from the Leibniz Institute for Natural Product Research and Infection Biology, Germany and his team had been working with&nbsp;<em>Psilocybe<\/em>&nbsp;<em>cubensis<\/em>&nbsp;for several years. Growing the mushrooms in their lab, they had seen the mysterious blueing reaction countless times. \u2018We were just curious and tried to solve a phenomenon that\u2019s been known for decades,\u2019 Hoffmeister says.<\/p>\n\n\n\n<p>But when they tried to extract and purify the blue compound, they failed. \u2018It puzzled and challenged us,\u2019 says Hoffmeister. \u2018This is where previous researchers \u2013 very talented people \u2013 had to give up, and that\u2019s where we went one step further with unconventional analytical methods.\u2019<\/p>\n\n\n\n<p>The researchers dug deep into the analytical toolbox with liquid chromatography\u2013mass spectrometry, Maldi mass spectrometry, infrared spectroscopy, as well as time-resolved nuclear magnetic resonance spectroscopy to observe the compounds as they form.<\/p>\n\n\n\n<p>The pigment, as it turns out, is not just a single compound but a complex mixture of linked psilocybin oxidation products. Most of them are quinoid psilocyl oligomers \u2013 compounds not unlike indigo, a deep blue pigment used to dye jeans. \u2018[The blue compounds and indigo] share structural similarities in the indole core, and in both the basis for the colour is a quinoid,\u2019 says the study\u2019s lead author&nbsp;<a href=\"https:\/\/www.leibniz-hki.de\/en\/institut-staff-details.html?member=918\">Claudius Lenz<\/a>.<\/p>\n\n\n\n<p>All of the six mushroom pigments the team identified are products of a cascade reaction starting with psilocybin. A phosphatase enzyme takes off its phosphate group, converting it into psilocin. An oxidising laccase then creates psilocyl radicals, which combine to form C-5 coupled subunits and then further polymerise via C-7. \u2018I think they did a beautiful job of showing the cascade reaction,\u2019 says&nbsp;<a href=\"https:\/\/pharmacy.utah.edu\/medchem\/faculty\/current-faculty\/Winter_J.php\">Jaclyn Winter<\/a>, who studies natural product biosynthesis in bacteria and fungi at the University of Utah, US.<\/p>\n\n\n\n<figure class=\"wp-block-image\"><a href=\"https:\/\/www.psychedelicsworld.org\/\"><img decoding=\"async\" src=\"https:\/\/d2cbg94ubxgsnp.cloudfront.net\/Pictures\/\/web\/w\/v\/r\/lenzetalscheme2_978257.svgz\" alt=\"A scheme showing the blueing reaction in P cubensis, catalyzed by PsiP and PsiL\"\/><\/a><\/figure>\n\n\n\n<p>Source:&nbsp;<a href=\"https:\/\/doi.org\/10.1002\/anie.201910175\">\u00a9 2019 Claudius Lenz et al<\/a><\/p>\n\n\n\n<p>Proposed reaction scheme for the conversion of psilocybin into blue pigments<\/p>\n\n\n\n<p>What exactly the blue pigments do, however, remains a mystery. \u2018Our hypothesis \u2013 and we don\u2019t have any evidence for this yet \u2013 is that it might serve a protective role, like an on-demand repellent against predators,\u2019 says Hoffmeister. The compounds might produce reactive oxygen species, which are toxic to any insect nibbling on the mushrooms. \u2018I think we\u2019re going to see a lot of follow-up studies on the true ecological role of these molecules,\u2019 Winter says.<\/p>\n\n\n\n<p>Hoffmeister hopes that his study not only inspires others to&nbsp;<a href=\"https:\/\/www.chemistryworld.com\/5970.article\">study fungi from a chemistry perspective<\/a>&nbsp;but also changes people\u2019s mind about psilocybin. \u2018Psilocybin is looked at as this illegal, recreational drug, but it has a fantastic potential as a medication for therapy resistant depression,\u2019 he says.<\/p>\n\n\n\n<p>Winter agrees. \u2018There\u2019s quite a few groups who are studying psilocybin, and especially because it\u2019s been legalised in the US in various states, and because it\u2019s in clinical trials,\u2019 she says. \u2018I think [this study] is going to have a huge impact in the field.\u2019<\/p>\n\n\n\n<p><a rel=\"noreferrer noopener\" href=\"https:\/\/www.facebook.com\/sharer\/sharer.php?u=https:\/\/www.psychedelicsworld.org\/why-magic-mushrooms-turn-blue\/\" target=\"_blank\"><\/a><\/p>\n","protected":false},"excerpt":{"rendered":"<p>Why do magic mushrooms turn blue when they are cut?
